The Expression of OPG and RANKL from the Macrophage after Sensitization with Titanium Particles

Abstract
Purpose
Macrophages are activated by the wear particles and induce osteolysis and loosening of the Total Hip Arthroplasty (THA). And we did in-vitro study to confirm whether Osteoprotegerin (OPG) and Receptor Activator of Nuclear Factor (NF)-B Ligand (RANKL) were expressed from the macrophages which were sensitized with titanium particles. Materials and Methods: We used mouse macrophage cell line, J774A.1, and we obtained the titanium particles from the interfacial membranes of the loosening Total Hip Arthroplasty (THA), and filtered them to collect particles smaller than 10 m. Then we applied these titanium particles into the macrophage culture media in 0.01%, 0.1% and 0.5% (wt/vol) concentration, did RT-PCR to identify their effect on the expression of OPG and RANKL.
Results
The expression of RANKL was the highest, 2.17, after one hour sensitizing with 0.01% titanium and then decreased. By 0.1% titanium, the expression of RANKL was peak, 3.00, after two hours, and 0.5% titanium was toxic to macrophages. But OPG was not expressed in any groups.
Conclusion
RANKL expression of the activated macrophages by titanium particles was elevated and may involve in the process of osteolysis of the THA.
