The Effect of G174C Polymorphism in the Promoter Region of Interleukin-6 Gene on Perimenopausal Bone Metabolism and Serum Osteoprotegerin Level in Apparently Healthy Korean Women

Abstract
Background
Interleukin-6 (IL-6) is a pleiotropic cytokine playing a central role in bone turnover. Recent studies demonstrate its important role in the bone loss after loss of sex hormones, such as, menopause or ovariectomy. We investigated the frequency of G174C polymophism in the promoter region of IL-6 gene in perimenopausal Korean women, and analyzed its association with bone metabolism and serum osteoprotegerin (OPG) levels.
Methods
In 268 Korean women (mean age 51.4 yrs, range 37~73 yrs), anthropometric measurements were done and lumbar spine and femoral neck BMD, bone turnover markers, such as alkaline phosphatase (ALP), serum calcium, phosphorus, 24hrs urine calcium, phosphorus excretion and urine deoxypyridinoline were measured. Serum follicular stimulating hormone (FSH) levels were measured and serum OPG levels were measured with sandwich enzyme-linked immunosorbent assay (ELISA) method. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was performed.
Results
The allele frequencies were 0.965 for G allele and 0.035 for C allele, which were in Hardy-Weinberg equilibrium (p=0.50). There were no differences in the mean values for bone turnover markers, serum osteoprogetein levels and bone mineral density (BMD) between those with or without the C allele (p>0.05). There were no differences in the prevalence of normal, osteopenia and osteoporosis among different genotypes (p>0.05). When the analyses were done separately in two groups divided by menopausal status, there were no differences in the mean values of BMD between the genotype groups (p>0.05).
Conclusion
G174C polymorphism in the promoter region of IL-6 gene is observed in similar frequency in Korean perimenopausal women compared with other Asian races. G174C polymorphism showed lack of association with BMD or serum osteoprotegerin levels, and the results didn't show any differences according to the menopausal status.
