Abstract |
The histomorphometry of undecalcified bone section is another special histology technique. Once the province of bone biologic for studying normal bone metabolism, it is now a useful diagnosis tool. With this procedure, specific metabolic bone diseases can be diagnosed and studied and their response to treatment evaluated. Using undecalcified bone, a computer-assisted analysis of at least 50 microscopic fields generates precise data about four major parameters of bone metabolism. First, the amount of bone, measured as cortical thickness and trabecular volume, is quantified. Second, the rate of bone turnover can be calculated from the amount of osteoblastic and osteoclastic activity. Third, the functional capacity of osteoblasts can be evaluated by measuring their bone formation rate. Fourth, the efficiency of mineralization as measured by the mineralization lag time, can be calculated.
